The EB-CLARIFY provides you with the ability to carry out multiple sequential experimental processing procedures all within the same culture microplate. Specifically, the microplate is designed for EB formation, spheroid growth, tissue clearing, immunolabeling and imaging. Unlike conventional 3D culture workflows which require transferring the contents between multiple vessels, there is no transfer or manipulation of the delicate spheroids using the EB-CLARIFY!
The unique design of the EB-CLARIFY is optimized to drastically decrease the risk of damaging the microtissues compared to conventional setups. Each experimental unit of the EB-CLARIFY consists of two adjacent wells positioned on different levels that are connected via microchannels. The higher-laying solution exchange chamber is exclusively used for adding and removing solutions that are necessary for processing the spheroids. The lower-laying spheroid chamber houses the 3D culture, which is completely safe from accidentally being removed from its well due to pipetting. The microfluidic channels between the two chambers limit the intensity of media changes and washing steps, protecting the integrity of delicate organoids. The design is optimized to allow for a disturbance-free yet time-efficient solution exchange in the spheroid chamber.
This next-generation microplate is fully adapted to the standard 96-well microplate format, and is compatible with automated lab equipment. Each microplate houses 48 spheroids and is optimized to safely employ automatic washing equipment and pinning tools. This time-saving setup will help to massively increase the efficiency in drug discovery, efficacy and safety testing using spheroids.
The microplate features an ultra-thin and optically clear PDMS base, which has low auto-fluorescence. High-resolution images of the generated spheroids can be recorded with ease using plate readers and high-content screening platforms. As for our other EB generation devices, the spheroid chamber features a conical shape enabling the highly reproducible growth of uniform spheroids right in the center of the well.
